In vitro culture is a vegetative propagation method that allows obtaining uniform seedlings, fast clonal multiplication and ideal for preserving genotypic characteristics. The objective of this work was to determine the influence of surface disinfection and culture media in each of the stages of in vitro culture (initiation, multiplication and rooting) for an accession and a stevia mutant. In surface disinfection, eight treatments were developed (3; 5; 10; 25 minutes of immersion in 1 % and 1,8 % NaClO). The consistency (liquid and solid) and concentration (50 % and 100 %) of salts of the MS medium as starting medium were evaluated. The type of container for propagation was also compared. In the in vitro multiplication stage, the influence of kinetin (0,3; 4 and 6 mg/L) seeded in solid media with 50 % and 100 % MS salts was determined. In the in vitro rooting stage, auxin was evaluated (0,5; 1 and 2 mg/L of IBA and 0,1; 0,2 and 0,5 mg/L of ANA). Finally, some morphological characteristics of the Miskibamba accession and a mutant were compared in the best medium obtained. For disinfection it was better to immerse for 3 minutes in 1,8 % NaClO. The chosen starting media were 50 % and 100 % solid MS salts. The use of the glass vials was ideal for in vitro propagation. The culture medium for in vitro multiplication and rooting was MS + 0,2 mg/L ANA, because it allowed obtaining seedlings with greater length, number of internodes, leaves and roots, and consequently a greater number of explants to subculture. The mutant seedlings had better length, leaves and root in MS medium without growth regulator, while the original in MS + 0,2 mg/L of ANA.
