The aim of this study was to evaluate the effect of different cryopreservation times (6 and 9 minutes) and two levels of cryoprotectants (Level 1: 10% DMSO at 0.1 M, and Level 2: 43% dextrose at 0.3 M) on sperm viability of Colossoma macropomum. Semen samples from two males were collected in 0.4 M solution of sucrose, using 25 mL of thawed semen for eggs fertilization process. Results obtained from post-thawed sperm showed no significant differences for sperm motility rate (p˃ 0.05), however those mean values were lower when compared to motility rate recorded for fresh sperm. Only eggs fertilized with sperm from Male 1 submitted to 9 minutes of cryopreservation showed significant differences for fertilization rate (p=0.000001). Hatching rate was not affected by cryopreservation time (p>0.05), but for Male 2, samples treated with 10% DMSO solution showed better hatching rate levels (p=0.043). Neither congelation times nor levels of cryoprotectants seemed to have a significant effect in the rate of larvae malformation (p>0.05) but values obtained from post-thawed sperm were higher when compared to those recorded for fresh sperm. Therefore it is evident that there is an unknown male factor, which prevented replicate the values obtained, which is manifested in the synergy of the variables studied, for this species.
