There is worldwide concern about the loss of cattle diversity, and Peru is no stranger to this problem, considering that there are no cattle conservation programs and there are few genetic characterization studies. At the same time, knowing the genotypic variants of a population is crucial because it allows us to know its characteristics to take management measures that guarantee food security and conservation. In this context, the present study aimed to genotypically characterize the dairy proteins αS1-casein and αS2-casein in Brown Swiss, Holstein, Creole cattle from Apurímac and Amazonas. The genotypes of the αs1-casein (CSN1S1*BB, CSN1S1*BC y CSN1S1*CC) y αs2-casein (CSN1S2*AA, CSN1S2*AB y CSN1S2*BB) proteins were determined in 361 vaccine DNA samples from the Bank of DNA from the Directorate of Resources and Biotechnology of INIA using high resolution dissociation curve assays (HRM, High Resolution Melting). The frequencies of the CSN1S1*B allele were 0.99, 0.92, 0.71 and 0.78 in Holstein, Brown Swiss, Apurímac creole cattle and Amazonas creole cattle, respectively. While the frequencies of the CSN1S2*A allele were 1.0, 0.99, 0.98 and 0.94 in Holstein, Brown Swiss, Apurímac creole cattle and Amazonas creole cattle, correlatively. The FST values in CSN1S1 indicated the existence of greater genetic variation among the Creole cattle compared to the Holstein cattle breed.
