Background: Urine culture is the confirmatory test for urinary tract infection - UTI. The required sample is sterile urine, which must be delivered for processing within 2 hours of collection. Therefore, its collection, conservation and transport are critical steps to obtain reliable results for UTI diagnosis. Objectives: Determine stability of urine samples collected for urine culture in sterile containers without preservative, and with preservative/additive. Materials and Methods: A longitudinal descriptive study was carried out on urine samples (n=221) collected in sterile wide-mouthed containers for urine culture. For the study, 2 aliquots were obtained upon arrival for evaluation, and kept in: Device A (urine container without additive) refrigerated (4 -8°C) and Device B (BD Vacutainer Plus Urine C&S Preservative Tube) at room temperature. The urine cultures were processed in parallel, at time intervals of 2, 4, 8, and 24 hours, following the standard procedures of Microbiology Department of Cayetano Heredia Hospital. Results: Devices A without additive in refrigeration, showed statistically significant changes after 2 hours of conservation; positive urine cultures for uropathogens increased progressively, negative urine cultures with no growth migrated towards contaminating and positive growth. In devices B with preservative kept at room temperature, the changes were minimal, without statistically significant variation in the number of positive and negative urine cultures during the study period. Conclusion: The use of devices with preservative additives maintained the stability of the urine sample for urine cultures for up to 24 hours at room temperature.
