The future of the adjuvants development for vaccines consists in the combination of two or more of them to obtain a powerful immune response in poultry. Resiquimod (R-848), a synthetic TLR-7/8 agonist, together with Adyuvax, an adjuvant based on calcium phosphates, Al and Mg salts, might have a synergistic effect. Therefore, the objective of this study was to evaluate the effect in vitro of the co-administration of Adyuvax with R-848 on the relative expression and production of proinflammatory cytokines in chicken macrophages infected with virus of Newcastle Disease. Using the MTS assay, the cytotoxicity of both adjuvants on HD-11 chicken macrophages was evaluated. Subsequently, the cells were stimulated with the Newcastle disease virus (vENC) and treated with doses of each adjuvant for 6, 12, 24 and 48 hours. The mRNA expression levels of IL-1β e IFN-γ were evaluated for each exposure time to the different Adyuvax and R-848 treatments by real-time PCR and cytokine production by ELISA. In parallel, iNOS gene expression and nitric oxide production were evaluated. It was shown that the combination of both adjuvants was not toxic to HD-11 cells. On the other hand, the combination of Adyuvax and R-848 showed similar results to the R-848 group regarding the relative expression of IL-1β, IFN-γ, iNOS and nitric oxide production in HD-11 cell line. The combination of Adyuvax and R-848 just presented a synergistic effect and with a statistically significant difference to the other treated groups in the relative expression of iNOS and IFN-g at 12 h and 6 h respectively, but it is not sustained over time. This effect is attributed to the immunomodulatory action of R-848. The integration of these two adjuvants in the formulation of inactivated vaccines can be useful to improve the cellular immune responde of inactivated vaccines in poultry.
