Porcine cysticercosis is a disease caused by the ingestion of Taenia solium eggs and their conversion intro oncospheres. Oncospheres penetrate the bloodstream disseminating in various tissues, The symptomatology depends on the place they deposit. Affected pigs often do not show symptoms due to their short lifespan before slaughter, so they can easily spread the disease by acting as intermediate hosts in the parasite’s life cycle. The diagnosis of porcine cysticercosis is based on the observation of the cysts during the necropsy of pig carcasses and through the palpation of the larvae at the base of the tongue, which are complicated tests to perform because they require trained staff. These tests are supported by serological tests such as immunoelectrotransferblot with affinity to lentil lectin glycoproteins (LL-GP EITB) due to its moderate sensitivity (77.37%) and specificity (76.4%) to diagnose it, however this is a test that depends on infected pigs to obtain the glycoproteic antigen. This is the reason we are looking for proteins that are easy to detect using serological tests and can be produced by modern methodologies such as recombinant techniques. TsAg5 has been detected in cyst fluid and excretion/secretion antigens of T. solium, and has been expressed in a prokaryotic system, without post-translational modifications, which showed a moderate sensitivity for detecting neurocysticercosis and a marginal enzyme activity. In this study, TsAg5 was expressed in insect cells, a system that provides post-translational modifications (e.g., glycosylation), antigenicity and correct folding, generating functional proteins with trypsin-like enzymatic activity. In addition, its antigenicity was verified with pigs’ sera with cysticercosis through western immunoblot.
