Early and specific diagnosis is one of the main objectives in malaria control strategies. The present study assesses Loop-mediated isothermal amplification (LAMP) technology, validating a commercial system and developing an in-house colorimetric LAMP system using multicopy gene sequences to detect Plasmodium vivax and Plasmodium falciparum with direct visualization of the color change of the neutral red indicator from yellow to fuchsia in the positives reactions. The limits of detection (LOD) of the test developed were estimated to be between 2.4 and 3.7 parasites/μl of blood. The best performance for the P. vivax detection was obtained with the COX1 amplification target, while the design with the Pfr364 amplification target was the most specific for P. falciparum detection. In the P. vivax and P. falciparum diagnosis, the results using whole blood sample indicate that the colorimetric LAMP in-house can detect between 36.6 and 80.2% of submicroscopic infections. The maximum detection capacity of colorimetric LAMP in-house was achieved using purified DNA with silica filter; however, a rapid sample preparation (Boil & Spin) showed result as reliable as those obtained by microscopy in samples of whole blood or blood impregnated on filter paper.
