MACOMCH-01 is a semi-synthetic macamide that has shown in-vitro inhibitory activity of fatty acid amide hydrolase (FAAH), an enzyme responsible for the metabolism of endocannabinoids. This fact suggests a potential antidepressant effect. However, to date no studies have been reported to verify this potential in preclinical models. The present study evaluated the effect of MACOMCH-01 administration at three different doses on depressive-like behavior in BALB/c mice. Likewise, the effect on endocannabinoid (Napepld, Faah, Cnr1 and Cnr2) and dopaminergic (Drd1 and Drd2) neurotransmission-related gene expression in the nucleus accumbens was also be determined. For this purpose, 97 male BALB/c mice, 2 months old were used. The forced swimming acute stress model was applied. Depressive-like behavior was evaluated using the same forced swimming test, while the anxiogenic and locomotor seconday effect were evaluated using the light-dark box and open field tests, respectively. For the analysis of the level of gene expression, midrodissections of the nucleus accumbens (core) and RNA relative quantification by RT-qPCR were performed. The dose of 10 mg/kg i.p. MACOMCH-01 reduced the depressive phenotype in male BALB/c mice exposed to acute stress. Likewise, MACOMCH-01 increased the expression of Faah and Cnr1 in the nucleus accumbens, while Drd1 and Drd2 did not show significant changes in the expression level.
