Norovirus is the leading cause of acute non-bacterial gastroenteritis worldwide, with the GII.4 genotype responsible for the majority of cases reported to date. Due to a lack of drug treatment, an early diagnosis takes an important role. Currently, serological methods for the detection of specific antibodies use virus-like particles, which represent a limit for laboratories in low-resource conditions. The present study aimed to design a multiepitope protein in silico for the detection of IgG and IgM antibodies against Norovirus GII.4 in human serum. A total of 20 immunodominant epitopes of the VP1 protein of the norovirus GII.4 capsid were predicted by immunoinformatic analysis, of which 10 were selected to construct the synthetic gene MEP-NVGII.4. This recombinant gene was expressed in E. coli BL21, and the recombinant protein, MEP-NVGII.4, purified and used as an antigen for the development of an indirect ELISA for the detection of IgG and IgM in a group of 26 sera of babies aged between zero and 2 years. Similar values were observed between serum IgG levels of infected individuals and controls. However, a significantly high statistical difference (p <0.0001) was obtained when comparing serum IgM levels. MEP-NVGII.4 demonstrated a potential utility for seroprevalence studies as a diagnosis of acute and convalescent infection with norovirus GII.4.
