Aeromonas salmonicida and Yersinia ruckeri are pathogenic bacteria of rainbow trout; Its early detection through the use of diagnostic tests is essential to avoid bacterial spread and excessive mortality in production systems. However, the use of diagnostic tests are not implements because they are expensive, require sophisticated laboratories and are carried out in cities far from production centers. Therefore, this work aimed to standardize the Loop Mediated Isothermal Amplification (LAMP) technique, a highly sensitive and inexpensive rapid molecular diagnostic test, to simultaneously detect A. salmonicida and Y. ruckeri in isolated strains of rainbow trout. LAMP primers were designed for the detection of A. salmonicida and Y. ruckeri through the vapA and rucC-rupB genes, respectively. A LAMP DUPLEX protocol was developed, running at 60°C for 60 min and stopped at 80°C for 10 min. The primers were specific for A. salmonicida and Y. ruckeri, not observing cross-reactions with Aeromonas hydrophila, Infectious Pancreatic Necrosis Virus, Flavobacterium psychrophilum, Renibacterium salmoninarum, species related to infectious diseases of rainbow trout. In addition, Staphylococcus aureus, Echerenchia coli, Pseudomonas aeruginosa and Yersinia enterocolitica, bacterial species related to environmental contamination and handling. Finally, the analytical sensitivity of LAMP was superior to the gold standard PCR technique.
