Introduction: Hydroxyapatite is the main mineral component of bone tissue in humans. There are multiple studies in the literature on obtaining hydroxyapatite from natural residues of animal, aquatic, coral and eggshell origin. Objective: To develop an experimental bone filler matrix based on nanohydroxyapatite obtained from chicken eggshell with silver nanoparticles. To assess newly formed bone density in alveoli treated with this experimental bone filler matrix using bone histomorphometry in an animal model. Material and Methods: Nanohydroxyapatite was synthesized from chicken eggshells under the sol-gel method and nanoparticles by the chemical coprecipitation method. The characterization of this experimental matrix was given by the X-ray diffraction technique. Cytotoxicity was assessed by the CC50 test in Madin Darby Canine Kidney cell lines and antibiotic susceptibility against Streptococco gordonii, Fusobacterium nucleatum and Porphyromonas gingivalis. To evaluate new bone formation, 15 Holtzman albino rats were assessed. The upper right and left first molars were extracted. The animal models were classified into 3 study groups: G1: commercial synthetic Hydroxyapatite GenPhos HA TCP-Baumer, Sao Paulo, Brazil; G2: the experimental bone filler matrix based on nanohydroxyapatite taken from eggshell of hen alone; and G3: the experimental bone filler matrix based on nanohydroxyapatite taken from chicken eggshell plus silver nanoparticles was grafted. To determine bone neoformation, a histomorphometric analysis was performed using images of the bone tissue of the alveolus at 30 days, which were delimited and processed in the IMAGE J program. Results: Nanohydroxyapatite was synthesized from chicken eggshell and silver nanoparticles, crystalline phases were identified with an ordered and periodic structure. The cytotoxicity test shows that the experimental matrix of nanohydroxyapatite plus silver nanoparticles reaches its cc50 at 93mg / ml, proving not to be toxic to cells. The antibiotic susceptibility test showed that the experimental matrix has an antibacterial effect on the Streptococco gordonii and fusobacterium nucleatum strains. Finally, the experimental matrix showed greater new bone formation 30 days after grafting compared to a commercial synthetic matrix (p = 0.031). Conclusion: The experimental filler of nanohydroxyapatite obtained from chicken eggshell plus silver nanoparticles proved to be non-toxic, exhibited an antimicrobial effect against S. gordonii and F. nucleatum strains, and promoted greater new bone formation.
