The aim of this study was to compare seminal characteristics of samples obtained by artificial vagina (AV) and by deviation of the vas deferens (DVD) processed with two different extenders (Steridyl® and Tris-lactose-egg yolk), the interaction effect was evaluated between both factors. Samples were collected from 6 adult male alpacas, 3 for the AV collection and 3 for the DVD collection. Vitality was assessed by Hoesch-Pi (Propidium Iodine) and functional membrane integrity and presence of acrosome were simultaneously assessed by the hypoosmotic test in combination with Coomassie Blue stain. Other characteristics like motility, sperm concentration and morphology were also evaluated. Post-thawing samples obtained by DVD and diluted with tris showed higher progressive motility (15.0 ± 7.5%) and Steridyl (15.6 ± 7.7%), whereas the lowest results were shown in samples obtained by AV diluted with tris (2.1 ± 3.4%) and Steridyl (2.9 ± 3.4%). Membrane integrity and vitality were superior in samples obtained by DVD diluted with tris (19.6 ± 11.7%; 31.4 ± 8.4%, respectively), in comparison with the results obtained by DVD diluted with Steridyl (12.1 ± 4.6%, 29.3 ± 9.7%) and with the results obtained by AV diluted with the same extender (12.1 ± 4.6%, 11.6 ± 4.2%). The lowest values were observed in samples collected by AV diluted with tris (9.8 ± 3.4%; 9.9 ± 3.9%). The results indicate that the samples obtained by DVD, either way diluted with tris or steridyl, showed the best results during the cryopreservation process in alpaca semen.
