Abstract: La investigación tiene como objetivo evaluar el efecto del bioisóstero Q7Cl del paracetamol sobre la nocicepción en Rattus norvegicus var. Holtzman. Para ello, se adquirieron 20 Rattus norvegicus var. Holtzman, las cuales fueron asignadas en 4 grupos de 5 especímenes cada uno; grupo control, patrón, problema 1 y problema 2, se administró por vía oral mediante jeringas de 1 mL; 0,5 mL de tween 80, 12,5 mg/kg de paracetamol; 12,5 mg/kg del bioisóstero Q7Cl y 25 mg/kg del bioisóstero Q7Cl, respectivamente. Posteriormente, fueron introducidas en el equipo de placa caliente a temperatura de 55°C, para medir el tiempo de respuesta al estímulo término mediante el lamido de la pata o el salto, los tiempos a evaluar fueron 30, 60, 120, 180, 240 y 300 minutos. Posterior al periodo de depuración de siete días, se procedió de la misma forma para el método de la placa fría con la diferencia que el estímulo térmico fue de 5°C. Los datos obtenidos fueron evaluados mediante el análisis de varianza (ANOVA) con el programa estadístico GraphPad Prism versión 8.0.2, al presentarse diferencia significativa se aplicó la prueba complementaria, prueba de Tukey. En placa caliente, el bioisóstero Q7Cl a la dosis de 12,5 mg/kg demostró un ligero aumento en el efecto antinociceptivo que el paracetamol (p>0,05) pero mayor efecto antinociceptivo estadísticamente significativo en comparación con el tween 80 (p<0,001) y mayor efecto antinociceptivo estadísticamente significativo en comparación con la dosis de 25mg/kg (p<0,05). En placa fría, el bioisostero Q7Cl a la dosis de 12,5 mg/kg y 25 mg/kg demostraron mayores efectos antinociceptivos estadísticamente significativos (p<0,01) en comparación con el tween 80 y mayores efectos antinociceptivos en comparación con el paracetamol (p>0,05). En conclusión, el bioisóstero Q7Cl a ambas dosis (12,5 mg/kg y 25 mg/kg) demostró tener efecto antinociceptivo en Rattus norvegicus var. Holtzman en la prueba de placa caliente (55 °C) y en la de placa fría (5° C)
ABSTRACT The research aim is evaluate the effect of the Q7Cl bioisostere of paracetamol on nociception in Rattus norvegicus var. Holtzman. For this purpose, 20 Rattus norvegicus var. Holtzman were acquired and assigned into 4 groups of 5 specimens each; control group, standard, problem 1, and problem 2, were administered orally by 1 mL syringes; 0.5 mL of tween 80, 12.5 mg/kg bw of paracetamol; 12.5 mg/kg bw of Q7Cl bioisostere and 25 mg/kg bw of Q7Cl bioisostere, respectively. Subsequently, they were introduced into the hot plate equipment at a temperature of 55°C, to measure the response time to the term stimulus by paw licking or jumping, the times to be evaluated were 30, 60, 120, 180, 240, and 300 minutes. After the seven-day depuration period, the same procedure was followed for the cold plate method, with the difference that the thermal stimulus was 5°C. The data obtained were evaluated by analysis of variance (ANOVA) with the statistical program GraphPad Prism version 8.0.2, and when a significant difference was found, the complementary test, Tukey's test, was applied. In the hot plate, bioisostere Q7Cl at a dose of 12.5 mg/kg showed slightly better antinociceptive effect as paracetamol (p>0.05) but increased antinociceptive effect statistically significant in comparison with tween 80 (p<0.001) and increased antinociceptive effect statistically significant in comparison with the dose of 25mg/kg (p<0.05).
