Since the beginning of the pandemic, the staff at the Regional Hospital of Ayacucho (HRA) has worked tirelessly for the benefit of the population, facing SARS-CoV-2 and being exposed to the virus more than the general population. This study aimed to determine the frequency of SARS-CoV-2 positivity among the HRA staff from January to April 2022. The study was descriptive-correlational and included 1,186 HRA workers. It followed a cross-sectional observational methodological design, collecting information without altering the environment. This allowed for estimating the frequency of SARS-CoV-2 at a specific moment and studying the association between demographic and labor variables with virus positivity using the Chi-square test (P<0.005) and calculating the odds ratio (OR) to quantify the magnitude of the association. The frequency of SARS-CoV-2 positivity was 33.38% among the surveyed workers. A higher positivity rate was observed in female staff (78.8%) compared to male staff (21.2%). The most affected occupational group was the healthcare area, with 90.9%, showing a statistically significant association with SARS-CoV-2 positivity compared to the non-healthcare area. Additionally, 23.5% of the positives had risk conditions, while 76.5% did not. The demographic and labor variables studied did not show a significant association with the frequency of SARS-CoV-2 positivity
