ABSTRACT The aim of this research report was to validate a high-performance liquid chromatographic method for the titration of Benzalkonium Chloride in an ophthalmic solution, for which placebo of Levofloxacin 5 mg/mL + Dexamethasone 1 mg/mL Ophthalmic Solution was mainly used. A method of quantification of Benzalkonium chloride was performed by means of preparations of a buffer solution, a diluent, a mobile phase and a standard by which parameters such as specificity, linearity, accuracy, precision and robustness were evaluated, following the recommendations of the United States Pharmacopoeia (USP). The method proved to be specific as there was no interference from other components in the analysis of Benzalkonium Chloride. In addition, a linear relationship between analyte concentration and method response was established, as it showed a correlation coefficient of r = 0.9991. Adequate accuracy was obtained with an average recovery of 100.31% within the required specifications. Furthermore, the precision of the method was evidenced by relative standard deviation values below the established limit (DRS<2%). The robustness of the method was verified by meeting the established acceptance criteria in terms of acceptable variation and coefficient of variation. The results obtained demonstrated the consistency, reliability, and reproducibility of the method, complying with good laboratory practice; the method was precise, consistent and accurate in the measurements, which guarantees reliable repeatability of the results under various conditions. In conclusion, the performed high performance liquid chromatography method was found to be specific, linear, accurate and robust for the titration of Benzalkonium Chloride in ophthalmic solution. These findings supported the reliability and quality of the method, allowing its implementation in pharmaceutical practice to ensure accurate and reliable titration of this critical component.
