ABSTRACT The objective of this study was to evaluate the effect of the freeze-dried extract of Juglans neotropica Diels on anxiety, depression and memory induced by maternal separation in Rattus norvegicus var. Holtzman. The botanical material was collected from the district of Chimbote, province of Santa, Ancash region, Peru, stabilized at -70ºC and then freeze- dried. 24 specimens of Rattus norvegicus var. Holtzman were used, divided into 2 groups, problem (subjected to maternal separation for 4 hours during 14 consecutive days) and negative control. To obtain the baseline data, the specimens were expected to reach adulthood (2 months). Four pharmacological models were used: elevated plus maze and open field to determine anxiety, Porsolt forced swim to determine depression and Morris test to later determine the negative alteration on memory resulting from the stress induced by maternal separation. After that, the Problem group was subdivided into 5 groups, Problem 1 (0.5 g / Kg) and Problem 2 (1 g / Kg) to which the lyophilized extract of Juglans neotropica Diels was administered for 14 days; Pattern 1 (bromazepam 10 mg / mL for 14 days), Pattern 2 (fluoxetine 1 mg / mL for 14 days) and positive control (physiological saline solution). Finally, the same pharmacological models were performed again to obtain the post-treatment data. The data were analyzed using the ANOVA test and Tukey's test as a post hoc test. The results showed that the 0.5 g / Kg and 1 g / Kg doses of the lyophilized extract of Juglans neotropica Diels presented a similar effect to bromazepam in the elevated plus maze and Open Field as an anxiolytic effect; Likewise, they presented a similar effect to fluoxetine in Porsolt forced swimming as an antidepressant effect, however, as an effect on memory impairment, no marked positive results were obtained with the doses of the lyophilized extract. It is concluded that Juglans neotropica Diels in doses of 0.5 g/kg and 1 g/kg has an antidepressant, anxiolytic effect in animal models and more studies are needed to corroborate the improvement effect on memory impairment.
