The research was basic, non-experimental design, correlational, retrospective cross-sectional type. The population consisted of 70 Covid-19 positive patients. It was found that there is an association between comorbidities, age and polypharmacotherapy. In the result according to the Chi-Square hypothesis test, comorbidity and age are associated with polypharmacotherapy. Where comorbidities (X2 = 6.296; p value = 0.012) and age (X2 = 6.090; p value = 0.014). According to Odds ratio comorbidity (OD 4.295; CI 1.313 - 14.043) and age (OD 3.675; CI 1.277 - 11.100); that is to say that patients with comorbidities are 4.295 times more likely to present polypharmacotherapy compared to those without comorbidity, likewise patients aged 60 years and older are 3.675 times more likely to present polypharmacotherapy compared to patients younger than 60 years
