In the blood, red blood cells for its metabolism consume the glucose of plasma or serum, generating inaccurate by the time spent from sample collection to processing. The objective was to standardize blood collection on filter paper and its treatment for glucose determination. Material and methods: From samples of volunteers we determined the area of graph paper, collection volume, drying time, concentration and volume of deproteinizing, agitation, temperature and centrifugation, and subsequently standardize the volume of deproteinized by the percentage of inhibition in the enzymatic method of glucose oxidase. Results: The appropriate paper was Whatman No 3, in an area of 100 mm2, the amount of blood of 20uL, the drying time 16 min. The deproteinized with 10% TCA with 200uL, without stirring, centrifugation or ice bath. The percentage of inhibition was 2.49% which is corrected by the factor of 1.075. Conclusion: The methods of blood collection on filter paper and treated with TCA for glucose determination is useful and valid with an r of 0.951.
