--- A basic requisite for any breeding program based in marker-assisted selection (MAS) is counting with a suitable number of polymorphic markers. Among the large variety of molecular markers available, microsatellites, short tandem repeat units of between 1 and 6 bp in length, offer appreciable advantages. Being a PCR-based technique, requiring only small amounts of DNA and their co-dominant nature have made them a very popular tool in animal and plant studies. Despite the importance of sweetpotato (Ipomoea batatas L.) in the world, only a small set of microsatellite markers are available for this crop. Aiming to generate a large number of SSR markers in sweetpotato, two genomic libraries have been constructed using DNA of an african sweetpotato cultivar “Tanzania“. In order to look for tri-nucleotide microsatellites, which frequently produce fewer “stutter bands” as compared to di-nucleotide microsatellites. The enrichment of genomic libraries achieved 1.59 % of a total number of 2900 clones evaluated by different screening methods as White/blue, Colony lift and Southern blot. Fifteen pair of primers were designed, and eight of them showed polymorphism in eleven sweetpotato accessions from different places of the world, which are kept in the germoplasm bank of the International Potato Center (CIP).
