Abstract: El objetivo del presente estudio fue determinar la expresión de péptidos
antimicrobianos (α y β defensinas) en el epitelio intestinal de crías de Alpaca con
enteropatías, de cero a 45 días de edad, mediante la cuantificación de ARN mensajeros
(ARNm) de las α (defa 8) y β (β def alp) defensinas, utilizándose oligoneuclótidos
diseñados por Patil y col. (2004) y a partir de secuencias publicadas en el Banco de Genes
(GenBank) para bovinos (Acceso: AF016539.1), respectivamente. Se tomaron porciones de
yeyuno, de 2 cm de longitud, de crías de Alpaca con signos de diarrea (n=29); además, se
realizó el análisis histopatológico del tejido utilizando la tinción Hematoxilina-Eosina para
determinar la enteropatía. Se obtuvo el ARNm total de los raspados yeyunales, que sirvió
de molde para la síntesis de cDNA mediante transcripción reversa (RT), seguido de un PCR
en Tiempo Real, para la amplificación y detección de las defensinas. La cuantificación
relativa de ARNm, se realizó mediante el método 2-ΔΔCt
. Las crías enfermas mostraron una
expresión de Defa 8 correspondiente a 18.15 veces lo expresado por el animal recién nacido
o calibrador. Similarmente, se observó que la expresión de β-def alp correspondiente a
258.29 veces lo expresado por el calibrador, encontrando diferencia significativa (p<0.05),
tanto en Defa 8 y β-def alp mediante la prueba T de Student, al compararlos con un grupo
de crías de Alpacas sanas criadas en similares condiciones de un estudio previo. Los
resultados demuestran que las α y β-defensinas entéricas son inducidas de manera
significativa durante cuadros entéricos en las alpacas.

--- The aim of this study was to determine the expression of antimicrobial peptides (α and β Defensins) in the intestinal epithelium of newborn alpacas from zero to 45 days old suffering enteropathy signs. We quantified messenger RNA (mRNA) of α (DEFA 8) and β (β def alp) defensins, using primers designed by Patil et al. (2004) and from published sequences in the gene bank (GenBank) for cattle complete mRNA (Access: AF016539.1), respectively. This was done by taking portions of jejunum, 2 cm long, from young alpacas with diarrhea (n = 29) and performed histopathological analysis of this tissue using hematoxylin-eosin staining to corroborate enteropathy cases. Total mRNA was obtained from jejunal scrapings, which served as template for cDNA synthesis by reverse transcription (RT), followed by Real-Time PCR for amplification and detection of defensins. Relative quantitation of mRNA was performed using 2-ΔΔCt method. Expression of Defa8 in young alpacas suffering enteropathy was of 18.15 fold times was expressed by newborn animals or calibrators. Similarly, we observed that expression of β-def alp was 258.29 times fold what is expressed by the same calibrators, finding statistical difference (p <0.05) of both, Defa8 and β-Defa def alp, by means of Student's T test when compared to a group of young healthy young alpacas raised in similar conditions of a previous study. The results conclude that enteric α and β-defensins are induced significantly during enteric disease in alpacas.
