-- The present study had 3 experiments and the main objective were to evaluate the effect of two superoxide dismutase mimics (Tempo and Tempol) during semen sperm cryopreservation and vitrification on alpaca sperm function. In Experiment 1, vitrification of alpaca sperm was evaluated using extenders based on Tris solution and HTF, with different sugars as cryoprotectants (fructose, sucrose, and trehalose) in different concentrations (0.25, 0.5 y 1.0 M). Only few spermatozoa motile (<1%) and viable (<5%) were found in group HTF.In Experimental 2, six samples of fresh alpaca sperm were incubated during 3 hours at 37 °C on a medium based on Tris with polymorphonuclear activated by 100 mM PMA (phorbol myristate acetate) to induce ROS production. Sperm motility was significantly reduced (p< 0.05) when sperm were incubated on a medium con high levels of ROS (13.67%) in comparison with control group (22.42%). In Experiment 3, two superoxide dismutaseanalogues (antioxidants Tempo and Tempol) were evaluated during cryopreservation process on an extender based on skim milk and ethylene glycol. Addition of Tempo or Tempol result in higher (p< 0.05) sperm motility in comparison than control group (19.63% and 22.57% vs. 10.62%). Functional sperm membrane integrity were also higher (p<0.05) in groups Tempo (14.31%) and Tempol (13.08) than control group (10.38%). In the same way, DNA integrity washigher (p< 0.05) in groups Tempo (74.20%) and Tempol (83.76%) than control group (63.68%). Viability and acrosomal integrity were not affected by cryopreservation with superoxide dismutase analogues. We conclude that alpaca sperm incubation on PMN activated by PMA had a detrimental effect on alpaca sperm motility, and that motility, functional sperm membrane integrity and DNA integrity could be partially protected by administration of Tempo or Tempol as components of extender for cryopreservation alpaca semen. -- Key words: Sperm, alpaca, cryopreservation, ROS, antioxidant, tempo, tempol
