Lomas are seasonal ecosystems occurring in the Peruvian coast. Bostryx aguilari (Mollusca, Gastropoda, Orthalicidae) is an endemic land snail which lives in Lomas from Lima. Genomic information (for example mitochondrial genome) has become a key factor to develop both phylogenetic and conservation studies. The present work is an evaluation of the genomic evolution of B. aguilari. Land snails were collected and DNA was isolated using CTAB method. The mitochondrial markers both Cytochrome oxidase I (COI) and 16S rRNA as well as the rRNA nuclear markers were amplified and sequenced. The three markers were used for genomic characterization and the evaluation of genetic diversity. Phylogenetic reconstruction was carried out also using sequences of other nine Orthalicid species and ten species retrieved from the GenBank. The phylogenetic trees were constructed with Neighbor-Joining, Maximum Parsimony, Maximum Likelihood and Bayesian Inference. DNA was difficult to amplify probably due to pigments in the muscle tissue of the snail foot. From the 80 extractions, eleven sequences of 16S rRNA from different populations of B. aguilari were obtained, four of COI and four of nuclear rRNA. The genomic characterization showed similar values with other species of the Family Orthalicidae. Genetic diversity showed low values. B. aguilari showed high differentiation from other Orthalicids and conformed a natural group. B. aguilari is more related to B. consperus and S. versiocolor than to other members of the Family Orthalicidae. Surprisingly, the genus Bostryx showed a polyphyletic pattern. The COI marker is an efficient barcode for B. aguilari. Molecular and morphological evidence lead us to conclude a B. aguilari does not belong to the genus Bostryx. B. aguilari would be a new genus. It is necessary more studies including more species to solve the evolutionary relationships within the Family Orthalicidae.
