The main objective of this work was to locate genes and molecular markers of alpaca (Vicugna pacos) using the Fluorescent in situ Hybridization (FISH) technique. For which the following specific objectives were proposed: a) Standardization of a cell culture protocol for alpaca lymphocytes and; b) Location of candidate genes for fiber diameter and color characteristics, and molecular markers of single nucleotide polymorphism (SNP), to chromosomal regions using the FISH technique. To carry out the first part of this work, two cell culture protocols were evaluated from alpaca lymphocytes. To physically locate genes and PNSs to alpaca chromosomes, it was necessary to have access to a BAC-CHORI 246 library of the alpaca genome. Radioactive labeling of gene and SNP probes hybridized to representative filters identified 326 BAC clones that putatively corresponded to the genes to be mapped. As a result, a standardized protocol has been developed for obtaining metaphase cells from the culture of alpaca lymphocytes in a specialized laboratory of the Universidad Nacional Agraria la Molina (UNALM). In turn, it has been possible to localize the genes ALX3, CHD7, CTNNB1, COL1A1, DAB2IP, KRT15, KRTAP13-1, NCOA6, SOX9, TNFSF12, ZIC1, ZIC5 to chromosomes 9, 29, 16, 17, 4, 16, 1, 19, 16, 16, 1 and 14 respectively. It has also been possible to estimate the locations of the SNPs BovineHD0700019108, BovineHD0800012560 and BovineHD2000012425, and the linkage groups to which they belong, to chromosomes 29, 9 and 17, respectively.
