The high incidence of anemia in Peru is largely due to the low consumption of iron-containing food, mostly affecting children under five years of age and pregnant women. Therefore, it is extremely important to search for efficient strategies to help prevent and cure anemia in our country. Ferritin is a multimeric protein that stores thousands of ferric ions and exhibits high bioavailability. The purpose of this research was to generate ferritin variants that cause iron accumulation in yeast. First, a soybean ferritin variant was constructed, in which five amino acids were replaced by those found in pea ferritin. Additionally, a plasmid library was built that allows the production of soybean ferritin variants with random sequences in that five amino acid domain. The replacement of amino acids 204-208 of soybean ferritin by those present in pea ferritin (Pro204Leu, Gln205Glu, Leu206Met, Ala207Thr and Asp208His) was successfully achieved, using an in vivo cloning strategy. However, the changes affected both the structure and function of the hybrid ferritin, which allowed only poor yeast growth in medium containing 10 mM ferrous sulfate. On the other hand, the plasmid library construction was successfully implemented, which will allow the analysis of a large number of random ferritin variants that include random changes in amino acids 204 – 208.
