The incorporation of cholesterol-loaded cyclodextrins (CLC) represents a strategy to increase the sperm resistance to the cryopreservation process, improving the sperm survival rates and the quality of the semen after thawing with respect to the untreated one. The objective of this investigation was to determine the effect of the addition of cyclodextrin loaded with cholesterol on the cryopreservation of bovine semen in the Holstein Friesian breed. The study was carried out in the Laboratory of the National Semen Bank of the UNALM, using 4 Holstein Friesian bulls from 3 to 4 years of age. A total of 28 ejaculates were collected using artificial vagina (7 ejaculated by bull) and their macroscopic (color, volume, pH) and microscopic characteristics (motility, vitality, concentration, integrity and functionality of the spermatic membrane) were analyzed. Subsequently, a 1: 1 pre-dilution of the semen was performed and divided into 03 aliquots for the addition of CLC (T1: 0 mg, T2: 1.5 mg, CLC and T3: 2 mg of CLC / 120 million sperm). It was incubated for 15 minutes at 37 ° C; then the final dilution and immediate packaging of the semen in 0.5 ml straws (30x106 spermatozoa) was carried out, performing the stepped temperature decrease, reaching -196º C. It was stored in tanks of liquid nitrogen; After 7 days, the straws were thawed for evaluation. The DBCA was used with sub sampling to measure the effect of the CLC in the different treatments. It was determined that there was no significant difference in the refrigeration stage in the different treatments. Post-thawing results indicate an improvement in the different variables studied using CLC, the total sperm motility was 67.73%, 79.59% and 74.73% with T1, T2 and T3 respectively. Vitality with T1: 63.26%, T2: 75.84% and T3: 70.53%. HOST tested values in T1, T2 and T3 with 46.89%, 60.43% and 55.06% respectively.
