In this research factors alkaline extraction and enzymatic extraction assisted protein sacha inchi (Plukenetia volubilis L.) are optimized, from the defatted cake, taking advantage of its high protein content (61,98 % dry basis) and being used to optimize the response surface methodology (RSM). In the alkaline extraction method it was performed primarily a screening to determine the variables that affect protein extraction sacha inchi by using the completely randomized design (CRD) with factorial 2k with replication in the central point, where the most influential factors on the extraction yield of the protein (response variables), pH, temperature, raw material/solvent ratio and the concentration of NaCl was determined. It used a central composite design (CCD) in step optimization, resulting in the following optimum conditions: temperature of 54,2 °C, solvent/raw material ratio 41,8/1 (v/w) and NaCl concentration 1,65 M, under the conditions of pH 9,5 and 15 min time; obtaining a maximum 30,2 g yield of soluble/100 g of total protein (experimental value) protein. In the method of assisted enzymatic extraction, optimization is directly applied by CCD for protein extraction yield (maximize) and the degree of hydrolysis (minimize). Subsequently, a multiple response optimization was applied using the function of Derringer overall desirability, in order to obtain both responses simultaneously, resulting in optimal conditions for each factor Alcalase enzyme concentration of 5,62 % and the time extracting 40,49 min; under the conditions of pH 9,0, solvent/raw material ratio 50/1 (v/w) solvent and temperature of 55 °C material, obtaining a maximum 44,78 g yield of soluble/100 g of total protein and a minimum degree of hydrolysis of 7,86 % (experimental value). Having recovered approximately 48 % more protein relative to the alkaline extraction method.
