The production of colourants from five filamentous fungi (Penicillium mallochii LMBHP19, Penicillium maximae LMB-HP33, Penicillium mallochii LMB-HP37, Talaromyces brunneus LMB-HP43 and Talaromyces wortmannii LMB-HP14) was compared in two fermentation systems: Submerged (SF) and by Surface Adhesion (SAF), using Czapek medium. Solid-State Fermentation (SSF) and Biofilm Fermentation (BF) were included in the SAF system, using inert supports (horticultural perlite and polyester cloth, respectively). The colourant production yields were calculated by UV-Visible spectrophotometry, considering the colourant units (CU) produced regarding biomass formation (g). Differences in the production of colourants were observed in the three fermentation systems. From these five strains, LMB-HP37 and LMB-HP43 presented the highest production yields in the three fermentation systems, showing the SSF system the highest yield values (633.75 CU/g biomass and 1019.92 CU/g biomass, respectively). While in the strains LMB-HP33 and LMB-HP14 no colourant production was detected in this system. However, LMB-HP14 achieved the highest values in the BF system. The results showed that exists a significant effect from the type of fermentation system and the fungal strain in the colourant production. In addition, due to the close relationship between the polyketide pathways for colourants and mycotoxins production, the expression of pks genes was evaluated in LMB-HP37 and LMBHP43. Although the presence of a pks gene was detected in the genome of both strains, only the expression of this gene in the LMB-HP37 strain in SF was detected by semi-quantitative RT-PCR. Nevertheless, this result would not necessarily imply the production of ochratoxins by this strain, since the pks gene is not unique to the synthesis pathway of these compounds.
