Cedroncillo (Aloysia herrerae) and pampa muña (Hedeoma mandoniana) are wild plants native to the Andes, which have been traditionally employed in traditional medicine. However, their potential as valuable sources of phenolic compounds has been underappreciated. This study explores the application of green technologies, specifically ultrasound-assisted extraction (UAE), known for its secure and economical high-yield extraction capabilities. The primary objective is to optimize the extraction of Total Phenolic Compounds (TPC) and antioxidant capacity (ABTS and ORAC) from cedroncillo and pampa muña leaves through UAE. Additionally, the optimal extracts are characterized using UPLC-PDA chromatography, and their biological activities are evaluated. The optimization process utilizes Response Surface Methodology (RSM) based on the Box Behnken statistical design. Independent variables investigated include time (5-20 min), temperature (30-70 °C), and ethanol concentration (30-80 percent). Estimated values for pampa muña indicate TPC (51.4 mg GAE/g dw), ABTS (426.2 μmol TE/g dw), and ORAC (1,524.0 μmol TE/g dw) under optimal conditions of 5.1 min time, 69.7 °C temperature, and 44.9 percent ethanol concentration. Correspondingly, cedroncillo estimates are TPC (91.4 mg GAE/g dw), ABTS (632.4 μmol TE/g dw), and ORAC (2,643.1 μmol TE/g dw) under optimal conditions of 12.3 min time, 61 °C temperature, and 57.3 percent ethanol concentration. The estimated results align with experimental outcomes, validating the mathematical models. Furthermore, optimal extracts from pampa muña and cedroncillo leaves were demonstrated through UPLC-PDA chromatography to contain predominant compounds derived from hydroxycinnamic acids and flavonoids, suggesting a promising potential for inhibiting enzymes such as α-amylase, α-glucosidase, pancreatic lipase, and angiotensin-converting enzyme (ACE). The pampa muña and cedroncillo extracts emerge as a promising alternative for various applications in the food industry.
