The objective of this work was to generate interspecific sexual hybrids resistant to P. infestans by means of unilateral sexual polyploidization of fourteen advanced tetraploid potato clones and fourteen interspecific diploid hybrids from the groups Piurana (Solanum chiquidenum and S. paucisecctum), Megistracroloba (S. sogarandinum) y Tuberosa (S. cajamarquense). From these crosses, a total of 1812 hybrids, grouped in 64 families were generated. Screening for late blight resistance was carried out under field conditions at Monobamba in 2012 using an alpha lattice design, 357 hybrids were selected by moderate to-high resistance levels to late blight. However, the well-documented positive effect of sexual tetraploidization in the resistance of the new hybrids seems to be accompanied by a worsening of pollen fertility. Only, 186 hybrids showed moderate to high pollen, and can be used as male parent in future crosses. The previous selected hybrids were characterized for their agronomic traits in field at La Molina 2013. Traits of number and length of stolons and yield shows statistically significant differences among cross combinations. Also, ploidy levels were estimated by counting the number of chloroplasts. Congruent with the frequency of 2n pollen in the male parents, the 91% of the hybrids were identified as tetraploids. Simultaneously, detached leaf assay with two complex races of P. infestans, PCO055 and PCO002 were performed to evaluate late blight reactions of hybrids selected, and to supplement large scale field testing for resistance to late blight. Finally, to verify late blight resistant levels, 231 hybrids were assessing under field infection at Monobamba in 2014, selecting 155 clones with proven moderate to high resistance. This is the first report of successful use of distant wild relative species from groups mentioned above for introgression of late blight resistance in the genetic background cultivated using unilateral sexual polyploidization.
