The objective of the present research was to evaluate the optimal conditions for the extraction of proteins from pajuro seeds (Erythrina edulis Triana). The optimization was carried out in three phases. The first phase was to screen six factors (time, agitation speed, temperature, flour/solvent ratio (MP/S), pH and NaCl concentration) using a 2k-3 fractional design (k=6) with aimed of evaluating their effects on protein extraction expressed in g soluble protein/g total protein. Being three the significant factors: time, temperature and MP/S ratio. The second phase consisted of optimizing protein extraction by using Box-Behnken design was applied which was generated by a second order model with a coefficient of determination R 2= 0.9976. The optimal conditions obtained for the MP/S ratio, temperature and time were 1/49 g/mL, 23 °C and 16 min, respectively. In the third phase, the model obtained with the Response Surface Methodology (MSR) was validated. The percentage of soluble protein recovery was 92.70, showing no significant differences with respect to the theoretical value (92.87), likewise, the protein yield of the process was 12.22 percent. Finally, characterization tests were carried out on the protein isolate obtained, whose results were: foaming capacity, 103.33 percent at pH 8; maximum foam stability, 60.88 percent at pH 6; water holding capacity, 3.53 g H2O/g isolate at pH 8; maximum solubility, 43.64 percent at pH 8; oil absorption capacity, 2.4 g oil/g isolate; emulsification capacity, 68.67 percent; emulsification stability, 91.30 percent and whiteness index, 75.35 percent. The optimization conditions obtained were favorable, obtaining a protein isolate with 92.45 percent purity.
