The treatment of sewage sludge in Wastewater Treatment Plants (WWTPs) is crucial globally due to the presence of pathogenic microorganisms and contaminants such as Salmonella spp., E. coli, viruses, parasites, and heavy metals. In this context, the legal framework, the Official Mexican Standard NOM-004-SEMARNAT-2002, aims to reduce environmental impact and protect public health. The detection of Salmonella spp. in sewage sludge is crucial, and traditional methods such as microbiological cultures have limitations. The use of molecular tests such as PCR is suggested. In this context, the hilA gene is of interest due to its association with bacterial virulence in Salmonella spp. strains. PCR of the hilA gene allows the study of this gene in biological samples, offering an effective alternative to traditional methods. In this work, PCR of the hilA gene was used to detect Salmonella spp. in sewage sludge samples, aiming for faster and more efficient detection than conventional microbiological and biochemical tests. The results showed that microbiological tests, following procedures established by NOM- 004-SEMARNAT-2002, allow for a preliminary identification of Salmonella spp. in sewage sludge. On the other hand, PCR of the hilA gene showed that it is not specific due to its presence in various bacteria. The possibility of errors in the specificity of this technique could be attributed to potential cross-contamination; however, the possibility of a horizontal gene transfer mechanism is not ruled out, since the matrix where these bacteria were found is dynamic and stressful environments, such as sewage sludge.
