Lepidium meyenii is a plant with various beneficial biological properties, with the black variety being most associated with male reproductive health. Evidence indicates that the administration of black maca in experimental animals improves seminal quality after altering spermatogenesis with lead; however, the physiological and molecular mechanisms explaining the effects of black maca on male fertility in a lead-induced oxidative stress model have not yet been elucidated. Objective: To determine if the antioxidant action of the aqueous extract of black maca protects against the decrease in spermatogenesis induced by lead acetate exposure in adult rats. This project contributes to laying the groundwork for proposing possible prophylactic approaches against male infertility and allows for future clinical research in humans exposed to lead. Methodology: 36 male rats were organized into 6 groups: the control group was injected with sodium acetate; Lead 8 and Lead 16 groups were injected with 8 mg/kg and 16 mg/kg of lead acetate (PbAc), respectively. The BM+Lead 8 and BM+Lead 16 groups were concomitantly administered 50 mg/day of aqueous black maca extract with lead acetate at 8 mg/kg and 16 mg/kg, respectively. The BM group was only administered 50 mg/day of aqueous maca extract. Various tests were performed, such as sperm count in the epididymis and vas deferens, and testicular histology was evaluated through histological sections. Additionally, the degree of oxidative stress and antioxidant effect were observed using the TBARS technique in testes. Results: According to the experimental design, it is determined that black maca at a dose of 50 mg/day significantly increases the sperm count in epididymis and vas deferens when comparing the group that was only administered PbAc at a high dose (Lead 16) with the group that was administered maca plus PbAc at the same dose (BM+Lead 16). Furthermore, histology showed that maca managed to increase sperm storage in the epididymis and improved tissue structure in the testicle. Additionally, black maca has not been found to significantly decrease MDA in the testicle. Conclusions: Black maca protects spermatogenesis from lead acetate toxicity in male reproductive organs, however, not through an antioxidant mechanism.
