Title: Caracterización morfológica y molecular de diferentes genotipos de Coffea canephora y el comportamiento en vivero de su progenie en Chanchamayo

Abstract: The present study was carried out with the objectives of morphologically characterizing and molecularly characterizing different genotypes of Coffea canephora and evaluating the behavior of their progeny located in the Germplasm Bank and Nursery of the IRD “Fundo Génova” in Chanchamayo during the 2018-2019 campaign. For the morphological characterization, 26 quantitative variables and 17 qualitative variables were evaluated that were taken from minimum descriptors established for Coffea canephora, of which the mean, standard deviation, minimum, maximum and coefficient of variability were evaluated, among the most important results were noted the null presence of coffee rust in the different genotypes, as well as marked differences in some productive variables. In the molecular characterization, the DNA was extracted and 4 ISSR-type primers out of a total of 10 were effectively used to differentiate the genetic variability of the different genotypes of Coffea canephora, the results of the dendrogram show that there is a notorious genetic variability between individuals belonging to the same accession. Finally, for the analysis of the behavior in the nursery of the progeny of the mother genotypes, the germination percentage of 1100 seeds distributed at random was evaluated, of which 150 seedlings were picked in polyethylene bags, where the number of leaves was evaluated, plant height and stem diameter per plant. Subsequently, 93 seedlings were extracted for the measurement of fresh weight and dry weight. The average results suggest a germination percentage of 78.64%, a number of leaves of 7.48, a height of 9.31 cm, a stem diameter of 0.25 cm, a fresh weight of 2.54 gr and a dry weight per plant of 1.25 gr.

El presente estudio se realizó con los objetivos de caracterizar morfológicamente y caracterizar molecularmente diferentes genotipos de Coffea canephora y evaluar el comportamiento de su progenie ubicadas en el Banco de Germoplasma y Vivero del IRD “Fundo Génova” en Chanchamayo durante la campaña 2018-2019. Para la caracterización morfológica, se evaluaron 26 variables cuantitativas y 17 variables cualitativas que fueron tomadas de descriptores mínimos establecidos para Coffea canephora, de las cuales se evaluaron la media, desviación estándar, mínima, máxima y coeficiente de variabilidad, entre los resultados más importantes se notó la nula presencia de la roya de café en los diferentes genotipos, así como marcadas diferencias en algunas variables productivas. En la caracterización molecular, se extrajo el ADN y se utilizaron efectivamente 4 primers tipo ISSR de un total de 10 para diferenciar la variabilidad genética de los diferentes genotipos de Coffea canephora, los resultados del dendrograma demuestran que existe una notoria variabilidad genética entre individuos que pertenecen a una misma accesión. Finalmente, para el análisis del comportamiento en vivero de la progenie de los genotipos madre, se evaluó el porcentaje de germinación de 1100 semillas distribuidas al azar, de las cuales se repicaron 150 plántulas en bolsas de polietileno, donde se evaluó el número de hojas, la altura de planta y el diámetro de tallo por planta. Posteriormente, se extrajeron 93 plántulas para la medición del peso fresco y el peso seco. Los resultados promedio sugieren un porcentaje de germinación de 78.64%, un número de hojas de 7.48, una altura de 9.31 cm, un diámetro de tallo de 0.25 cm, un peso fresco por planta de 2.54 gr y un peso seco por planta de 1.25 gr.
