The purpose of this study was to determine the effect of the acid etching and microretentions in the adhesion of the glass fiber post cemented with RelyX U100 system. The research is quantitative and research design is experimental. Forty single-rooted human premolar teeth extracted recently within 8 months. The instrument used was the data collection sheet to measure the adhesion of glass fiber post cemented with RelyX UlOO system, validated by expert opinion and test ofvalidity and reliability. The tensile bond strength val u es of glass fiber posts cemented with Rely X U 100 system according to manufacturer's specifications had an average of 80.23 N with a standard deviation of 29.82, minimum value of 39.20 N and the maximum value of 141.12 N, and acid etching and microretentions had an average of 102.68 N, with a standard deviation of ±38.29 N, where the minimum value of 60.72 N and maximum value of 217.56 N. Test Levene's F was applied to determine equal variances, obtaining the significance of 0.636, concluding that the variances are homogeneous in both groups. To applying the statistical test "t" Student' s "t" with n1 + n2 - 2 degrees of freedom, showed significant difference p = 0.0450 (p<0.05)between measures of adhesion by traction of post cemented with RelyX UlOO system with manufacturer's specifications compared with the post cemented with acid etching and microretentions. We can say that the acid etching of dentin and microretentions in the post increase the adhesion of glass fiber post cemented with RelyX UlOO system.
