The main objective of this study was to determine the expression leve! of the gene encoding the enzyme L-Galactono-1,4-lactone dehydrogenase and its relation to the content ofvitamin e in M dubia. Leaves and ripe fruits were obtained from the Instituto Nacional de Innovación Agraria Germoplasm eollection. Vitamin e content was determined by High Performance Liquid ehromatography and the relative gene expression by real time polymerase chain reaction. Vitamin e content of fruits was variable (650-2529 mg vitamin e/100 g pulp). In addition, vitamin e content of the peel was greater than that contained in the pulp or leaves (F = 363, df= 2,p < 0.001). Also, MdGalLDH showed differential expression in the three tissues (F = 97, df = 2, p < 0.001). Gene expression was up to 3x greater in plants producing high level ofvitamin e than those producing low levels (Student t-test = 3.24, df= 4, p = 0.032). In conclusion, M dubia plants from the INIA germoplasm collection contained fruits with a wide variation in vitamin e content, due in part, to differential MdGalLDH expression.
