Chronic Spontaneous Urticaria (CSU) shows functional autoantibodies in 50% of patients, which induce the release of histamine from skin mast cells and circulating basophils. Histamine is released from intracytoplasmic granules; the granules' membrane displays the CD63 molecule, which becomes exposed on the surface of these cells upon activation, and thus this molecule can be measured as a surrogate for basophil activation. The disease's course varies daily, so patient perception-based questionnaires (Clinical Tests) like Urticaria Activity Score (UAS7), Urticaria Control Test (UCT), and Quality of Life Questionnaire (CU-Q2oL) are recommended. The aim of this study is to establish the relationship between CD63 expression and CSU clinical test. A prospective study was conducted, evaluating patients clinically, performing urticaria questionnaires (Clinical Test), and measuring CD63 expression through flow cytometry using cells from an atopic donor and patient and control sera. Readings were done on the FACSCanto II flow cytometer, and analysis was done with FACSDiva 6.0 software. CD63 expression was considered positive with a stimulation index (SI) ≥ 2. The sample included 32 patients with CSU, assessing CD63 expression and 32 sera from healthy children were included as controls. In this work, CD63 expression had no correlation with the three clinical tests; a weak correlation was found between CD63 expression with UAS7 and CU-Q2oL. Regarding the UAS7 and CU-Q2oL clinical tests, they presented a good correlation coefficient of 0.589 with p=0.0001CD63 Further prospective studies with more patients are recommended to confirm these findings.
