Plants have been extensively used for medicinal purposes. Peru is rich in plant species, including Hura crepitans, a tropical tree from the Euphorbiaceae family.. PHARMA-DEV Laboratory (Toulouse, France) has isolated by ultra-high potency liquid chromatography new daphnane esters from the latex. Among them, Huratoxina and Stelleralide J, reduce by 30% the cellular viability of caco-2 cells, a colorectal cancer cell line, without affecting the control. Daphnane esters have been previously described as having different biological activities which include antitumoral ones, thus there is a great interest in exploring these compounds. This work used zebrafish embryos (Danio rerio) to study the effect of these compounds at organism level in aspects such as toxicology and biological activity. The lethal medium concentration (LC50) at 72 hours post fertilization (hpf) for Huratoxina is 1.60 μM while for Stelleralide J is 13.86 μM. The exposition of high compounds concentration (2-4 μM Huratoxina, 20-50 μM Stelleralide J) caused damaged in the envelope cells (cEVL) at 4 hpf. Immunostaining coupled with confocal microscopy of 6 hpf embryos revealed binucleated cells, phenomena previously reported for cellular division inhibitors. At low concentrations, both compounds caused a concentration dependent developmental delay. For Huratoxina (1-2 μM), cEVL damage seems to drag an effect throughout development resulting in undulated tail borders (a tissue originated by cEVL) at 48 hpf. Differently, Stelleralide J (5-10 μM) induces circulatory-hematopoyetic changes such as oedema formation at the caudal hematopoietic region, caudal and dorsal vessels flow blockage, blood flow reduction and decrease in neutrophils number. This preliminary study has identified the working concentrations of the compounds and biological effects important for cancer and other diseases treatment: cellular division and circulatory effects (angiogenesis), which justifies their posterior study in more specialized models.
