INTRODUCTION: The potency of anti-rabies vaccines for veterinary use produced by the National Institute of Health is determined using the NIH in vivo method, this biological assay has a high variability 25-400% due to many factors such as the animal's own response, its Environment, animal strain or line, virus strain for challenge test, analyst's expertise, inherent variability among others. The objective of the present study was to recognize certain factors related to the determination of potency. METHODS: It compiled information related to the potency test of the vaccines batch produced among 2011-2016. It compiled information related to the potency test of the vaccines batch produced among 2011-2016. The descriptive analysis of all the qualitative and quantitative variables was carried out, then the inferential analysis was carried out by the analysis of variance one way, where the potency of the vaccine and the antigen were evaluated with the other variables studied: pH and virus´s DL50. The analysis of variance of repeated measures was used to observe the behavior of the pH and the potency of the vaccine at different times of evaluation. The Splines method was used to determine the relationship between quantitative variables (vaccine and antigen potencies). Finally, it used the intraclass correlation method and the Bland and Altman method, to determine the degree of agreement between evaluators. The statistical programs Combistasts Demo version was used to carry out the evaluation of validity test, Stata 12 was used to the descriptive and inferential analysis (One-way Anova, Repeated measures Anova, Splines), the statistical tables were elaborated with Excel 2013, and the Epidat 4.1 program was used to determine the intraclass correlation. RESULTS: The results showed that the variability of the vaccine potency test was 24.7%, with a mean of 1.7 ± 0.4 IU / dose. The variability of the antigen was 26.3%, with a mean of 1.9 ± 0.5 IU / dose. A statistically significant difference was found between the antigen´s potency versus the different antigen´s pH groups (p <0.05). The Splines model showed that a segment of the antigen´s potency was significant explaining the vaccine potency variable, but as a total model it was not significant. The degree of agreement among the evaluators was moderate (67%). To evaluate the potency of the vaccine at different times, it was not statistically different in the months evaluated, but the pH changed significantly through the time. CONCLUSIONS: It was determined that there is a statistically significant difference among the different pH groups of the antigen with respect to their potency. It is also concluding that the pH of the vaccine varies over time.
