Parkinsonism is a clinical syndrome characterized by the presence of motor symptoms such as bradykinesia, resting tremor, and rigidity. This study sought to examine the effects of synthetic macamide (9Z, 12Z)-N-benzyloctadeca-9,12-dienamide administration at three different doses on parkinsonian behavior in rats, hypothesizing that synthetic macamide acts as a motor symptom reducer and FAAH inhibitor. Thirty two and a half months old Sprague Dawley rats were used and parkinsonism was generated by the antipsychotic haloperidol. Motor behavior was assessed by the open field test, catalepsy and the wire suspension test. The results of the open-field and catalepsy tests confirmed the haloperidol model. The catalepsy test indicated a delay in the cataleptic effect of haloperidol in the short term (30 minutes) of the 1mg/kg dose and in the medium term (90 minutes) of the 0.1mg/kg and 10mg/kg doses. The wire suspension test indicated decreased muscle strength in macamide administered rats. It is concluded that synthetic macamide (9Z, 12Z)-N-benzyloctadeca-9,12-dienamide delays the cataleptic effect and decreases muscle strength in rats induced to parkinsonian symptoms by an acute haloperidol model.
