Introduction: Platelet-rich plasma (PRP) is a therapeutic product, with a wide application in regenerative and cosmetic medicine, because it is an important reservoir of growth factors such as vascular endothelial growth factor (VEGF), responsible for the processes in angiogenesis and regeneration of damaged tissues. However, obtaining PRP does not present a current standardization, nor assessment of the different growth factors, affecting the effectiveness, time and money in the treatment of patients. General objective: To compare the amounts of VEGF released in activated PRP with different Glu.Ca dilutions, in PRP obtained by the Simple Centrifugation (CS) and Double Centrifugation (CD) methods. Materials and methods: 20 healthy volunteers between 18 and 40 years of age were enrolled. The blood samples were subjected to two centrifugation methods: CS and CD, the PRP obtained was activated with different dilutions and concentrations of Glu.Ca 10% 1/5 D1 (0.02 g/mL), 1/10 D2 (0.01 g /mL) and 1/20 D3 (0.005 g/mL), the released VEGF was quantified by direct ELISA at 450nm. Results: Concentrated platelets using the CS and CD methods showed a median and interquartile range of: 408.5 x 103 platelets/uL (382.75-473.0) and 532.5 x 103 platelets/uL (410.5-613.0), respectively. difference means between both methods. Regarding the VEGF quantification, a significant difference was found between the CS vs CD method for VEGF release with p<0.0001. Regarding the dilutions, no significant differences were found between the D1, D2 and D3 dilutions in the CS and CD methods with a p>0.05. Conclusions: The CS method showed a greater release of VEGF compared to the CD method. The Glu.Ca dilutions 1/5 D1 (0.02 g/mL) and 1/10 D2 (0.01 g/mL), were the ones that produced the highest release of VEGF in the CS method.
