The respiratory disease COVID-19 is caused by the coronavirus virus (SARS-CoV-2). At the ned of 2019, the first cases were recorded, which were detected in Hubei in China. As it is highly contagious, the World Health Organization (WHO) declared a global pandemic on March 11, 2020. For the reliable diagnosis of SARS-CoV-2, the real-time reverse transcription polymerase chain reaction (qRT-PCR) molecular test was used. The objective of this work was to evaluate the frequency of the E, N, RdRP genes detected in the diagnosis of SARS-CoV-2 with qRT-PCR during the period August-September-October 2020. The study was observational, descriptive, cross-sectional and retrospective. The data was taken from the 'Allplex™ 2019-nCoV Assay' Seegene program in the department of Lima. The number of cases is compared with the variables months, sex and age range by the frequency of genes confirmed for SARS-CoV-2. A significance value of p<0.05 is considered. A total of 13,360 confirmed SARS-CoV-2 cases were reported where the highest concentration of cases was in the month of August with 43.0% (5,742), September 39.8% (5,314) and the lowest October with 17.2% ( 2304). Confirmed cases by gender were 61.1% (8,158) for men and 38.9% (5,202) for women. Cases by age, the greatest range was between 30 to 39 years (19.6%), followed by the 20 to 29 years group (16.4%). 82.5% (11,019) of the cases were positive for the 3 genes E, RdRP, N. The mean obtained for the threshold cycle (Ct) was for the E gene Ct= 26.83, RdRP gene Ct=28, 73 and gene N Ct= 30.06. A significant relationship of p=0.018 was found between gene frequency and age. The N gene was the only one that presented a significant value p=0.007. No relevant association was discovered between sex and Ct value. It is also observed that the Ct value in those months was highly contagious. Therefore, it affected a percentage of the population's workforce. On the other hand, for the study of the clinical evolution of patients, the significance of the N gene will probably be a pillar
