This research work covered the preparation of Chitosan (CHT) films crosslinked with Sodium tripolyphosphate (TPP) and loaded with Chlorhexidine (CLX), which were obtained by the solvent evaporation method, also known as casting. The chitosan films (1%) were crosslinked with TPP (0.1%) using Glycerin (GLI) as plasticizer (10%) and loaded with CLX (0.12%) since these showed the desired mechanical characteristics to use them as a release system, such as to maintain its physical integrity after in vitro release. Films made of chitosan biopolymer (molecular weight 698.13 kDa, 71.04% degree of deacetylation) were characterized using different techniques to determine the chemical variations that take place in the films as a consequence of drug loading. Thus, the interaction with the polyanion TPP was evidenced by the FTIR technique, as well as the presence of the drug. Other characterization methods used were TGA and DSC to determine the thermal stability of the prepared films and SEM in order to observe the morphological variation of the film surface. The study of the degree of swelling was carried out to identify the behavior of the films in contact with the aqueous medium., where a second order kinetics was found. It is worth mentioning that the study of the release of Chlorhexidine was carried out at a controlled temperature (27 °C, 37 °C, 47 °C), the volume of gel used in the preparation of the film (25 mL, 35 mL, 45 mL) and the concentration of the crosslinker were varied (0.1%, 0.5%). Later, release profiles were adjusted to different mathematical models, where the Korsmeyer-Peppas model was the most appropriate to describe the release kinetics. Finally, the Bauer-Kirby diffusion test concluded that the films loaded with chlorhexidine showed fungicidal activity against Candida albicans, thus demonstrating that they work as a potential vehicle for drug administration since they allow the effective release of the active pharmaceutical ingredient.
