-- The purpose of this study was to evaluate in vivo antibacterial efficacy of 0.12% and 2% chlorhexidine gluconate (CHX) alternately whit 5.25% sodium hypochlorite (NaOCl) against anaerobic bacteria used as a root canal irrigating solution in teeth with pulp necrosis an radiographically visible periapical reaction. Eighteen single canals root were prepared using the step down technique. Nine canals were irrigated with 0.12% CHX alternately with 5.25% NaOCl, nine canals with 2% CHX alternately with 5.25% NaOCl. After accesing the canal, the first root canal sample was collected with a sterile paper point that were transferred to a tube containing reduced transported fluid (thioglicolate broth), the second root canal sample was following of irrigation . A small sterile cotton pellet was placed at the root canal entrance, and the cavity was sealed with zinc oxide-eugenol cement. The canals were maintained empty for 7 days; then one sterile paper point was introduced to absorb the root canal fluid (third sample). Samples were subjected to microbiologic processing, including anaerobic incubation on blood agar for 3 days. Finally the counting of CFU was calculated on the plates.
