The objective of this research was to choice the conditions that they permit to optimize the pectinasas enzymes production using orange skins and Actinomyces naeslundii; on first phase was used qualitative techniques for to determinate the conditions of pH, cultivation temperature, agitation and aeration for a good growth microorganism. On the second phase was used the Plackett- Burman design through the investigation of eight nutrients with two variation levels; The eight nutrients was: oranges skin, ammonium sulfate, urea, ferrous sulfate, calcium chloride, sodium chloride, magnesium sulfate y sodium carbonate. It was used Erlenmeyer flask with 150 mL of experimental medium pH 7, to 37 ºC, agitation to 300 rpm for 5 days. Third phase of optimization was developed through Box- Benhken design, oranges skin, ammonium sulfate and ferrous sulfate concentration were evaluated on three level. It was realized a multiple regression analyses with the three variables found previously . calculus and graphics were developed with Statistical 2.1 software. Optimum Parameters were: oranges skin 16.7 g / L, ammonium sulfate 5 g / L and ferrous sulfate 0.013 g / L. it was obtained 0.65 UI / mL of enzymatic activity in three days of biotransformation.
