Iron and copper are two micronutrients necessary for the generation of energy in our body and are found as structural and functional components of cytochrome c oxidase, located in the inner membrane of mitochondria, adenosine triphosphate production site to active skeletal muscle. Objective: Demonstrate isolate and simultaneous iron and copper deficiency diets decrease cytochrome c oxidase activity in skeletal muscle of rats. Research Design: Analytical, experimental, transversal and prospective. Methodology: Male Holtzman rats , 21 days of age, were used and randomly assigned in 4 groups of 7 animals to a control diet (containing 50mg of iron/kg of diet and 6mg of copper/kg of diet), an iron-deficient diet (containing 6mg of iron/kg of diet), a copper-deficient diet (1mg of copper/kg of diet), or an iron and copper deficient diet (containing 6mg of iron/kg of diet and 1mg of copper/kg of diet), respectively. The rats were fed for eight weeks, after a 14 hours fast, were anesthetize with chloroform. The blood samples were taken by cardiac puncture, and cuadriceps muscles were dissected out for measurement of cytochrome c oxidase activity. Muscle homogenates were prepared in order to isolate mitochondria by differential centrifugation and then the cytochrome c oxidase activity was assayed by measuring the decrease in optical density at 550 nm of reduced cytochrome c as it is oxidized by the enzyme following the method of Prohaska and Wells. Statistical analysis: The Kruskal-Wallis test was applied, then nonparametric Mann-Whitney test was used to compare medians. Program SPSS v 12.0 was used for data processing. Results: Statistically significant differences (p <0.05)in the cytochrome c oxidase activity of skeletal muscle were found, between the control diet group compared with copper deficient diet group (p = 0.006), control diet group compared with simultaneous iron and copper deficiency diet (p = 0.006), iron deficient diet group compared with copper deficient diet group (p = 0.009),and iron deficient diet group compared with simultaneous iron and copper deficient diet group (p = 0.013). Conclusion: The cytochrome c oxidase activity decreased significantly in skeletal muscle of rats fed with simultaneous iron and copper deficient diet and copper-deficient diet.
