--- In this work one Puno population sample was studied by the PCR (Polymerase Chain Reaction) amplification technique to detect the genetic variability of Y Chromosome STRs Markers DYS390, DYS391 and DYS392. These were compared with alleles frequencies with closest populations reported (Amerindian Population), the results show significant differences for the DYS392 STR. In addition two tables of population haplotype frequencies were constructed to compare South American populations with our Puno population and the other to compare with world-wide populations. The Puno population fits with the other peruvian populations studied (Arequipa and Tayacaja) keeping its distinction in South America. When compared with world-wide populations, the Puno population is similar with the Asiatic populations showing good correlation with the theories of human migration for peopling of the Americas as well as the conservation of genetic characters in this population.
