--- Lactose is the main carbohydrate in milk, their malabsorption is due to the nonpersistence of lactase enzyme, this deficiency or persistence can cause intolerance lactose. Non-persistence of lactase is variable in different populations around the world, this condition is related to the C allelic variant of LCT -13910 C/T polymorphism in the gene MCM6. The objective of the research was to establish the distribution of the frequencies of the polymorphism -13910 LCT/MCM6 gene in different Peruvians populations. The analysis was performed with 173 DNA samples from subpopulations of Lima (56.1%), Lima-Huarochirí (13.3%), Puno (17.3%) and Calca-Cusco (13.3%); the methodology consisted of genotyping by PCR- RFLP. The genotype frequencies in the overall sample were: C/C=98.8% and C/T=1.2%. In the subpopulations the results were: Lima, C/C=96.9% y C/T= 3.31%; Lima-Huarochirí and Cusco- Calca the C/C was 100%; Puno, C/C=98.3% and C/T=1.7%. Genotypes were in Hardy-Weinberg equilibrium, there was little genetic differentiation (Fst index). The allele frequencies, in the overall, were: C=98,8% and T=1,2%. In general, no differences for polymorphism LCT/MCM6 -13910 C/T in the peruvian subpopulations studied, with a high frequency of the genetic variant C, associated with lactase non-persistence and lactose intolerance. Keywords: Lactase/MCM6 gene, lactose, SNP -13910 C/T, genotype, allele, Peru.
