Introduction: Escherichia coli is a gram-negative bacterium that is transmitted through contaminated water, vectors (flies), and contaminated food. Antibiotics such as Ciprofloxacin are used for its treatment, while Kanamycin is used as a broad-spectrum antibiotic. On the other hand, some plants contain bioactive substances, such as phenolic compounds which are considered to have antimicrobial properties and beneficial health effects. There is evidence that when associating polyphenols with some antibiotics on bacterial strains, it is possible to observe both synergistic and antagonistic effects on cell growth. The aim of this research is to determine the relationship between caffeic acid, a hydroxycinnamic polyphenol, and the antibiotics ciprofloxacin and kanamycin on the growth of Escherichia coli. Materials and methods: We evaluated the relationship between caffeic acid-ciprofloxacin and caffeic acid-kanamycin against E. coli ATCC 25922 strain with the Checkerboard method. On the other hand, we determined the Minimum Inhibitory Concentration (MIC) of each compound using the serial microdilution method. Results: The MIC for caffeic acid, Ciprofloxacin, and Kanamycin were 2 mg/ml, 0.25 ug/ml, and 12.5 mg/ml, respectively. However, when caffeic acid is related to each of the antibiotics, an upward increase in the IC50 of each drug was observed, indicating a decrease in antibacterial action and as a result, an antagonistic effect. In addition, through the FIC index, antagonism was also determined, and that caffeic acid in a concentration of 1 mg/ml increased the MIC of Kanamycin and the MIC of Ciprofloxacin by approximately 4 times. Conclusions: Caffeic acid and antibiotics showed antibacterial effects when evaluated individually. However, by combining these substances, an antagonistic effect was observed. This effect is probably due to molecular competition between the compounds or to resistance mechanisms induced in the bacteria itself, such as the expression of efflux pumps or the recoding of metabolic pathways.
