Objective: To assess and synthesize the literature about the effectiveness and safety of the vaccines against the dengue virus. Methods: A systematic review of randomized clinical trials assessing the safety and efficacy of dengue vaccines was conducted. All clinical trials of CYD-TDV, TV 003 and TDV vaccines to determine efficacy, such as seroconversion according to the number of doses, dengue cases after the vaccine and number of hospitalizations, were included. The search was carried out in the databases of Medline, Cochrane library, LILACS, EMBASE and gray literature. The data extraction was carried out by two authors independently. We present summary measures that were used for efficacy and safety, such as relative ris (RR) or relative risk reduction (RRR) according to the intention-to-treat (ITT) and per protocol (PP) analysis, and for immunogenicity difference of means. Results: Six studies were included, four about CYD-TDV and two about TDV. The efficacy of CYD-TDV by ITT varies from 54.8% to 64.7%. The efficacy of TDV by ITT was 56% in the group that received two doses, 83% in the group that received a dose and 69% in the group that received a dose more reinforcement. Conclusion: The CYD-TDV and TDV vaccines have moderate efficacy. Efficacy according to seroprevalence prior to vaccination and circulating serotypes in endemic areas should be considered.
