Malaria in Peru is an endemic disease, which constitutes a public health problem, mainly in the department of Loreto, which concentrates high proportions of asymptomatic and submicroscopic infections. The objective of this work was to standardize a method of enrichment of genomic DNA of P. falciparum. DNA samples of P. falciparum of different parasitic concentrations were enriched through the selective whole genome amplification (SWGA) method, using the phi29 polymerase and the previously reported sets of primers 6A and 8A. Through SWGA, the initial DNA of P. falciparum in simulated clinical samples was increased to ~ 126 times (> 5 000 molecules/μL). The use of the FspEI enzyme generated an increase of ~ 38 times the DNA of P. falciparum and the total elimination of human DNA. The initial DNA of P. falciparum in clinical samples of patients with malaria was increased to ~ 29 126 times (> 60 000 molecules/μL). In conclusion, the SWGA method produces the enrichment of P. falciparum DNA in a wide range of concentrations, from simulated clinical samples and clinical samples of patients. Finally, this work provides a useful tool to the Malaria research groups in the Loreto region, and is an important element to carry out different studies that involve the DNA of samples of low parasitic density.
